I am Dr George Okechukwu Chukwuma, Virologist and Reader with the Department of Medical Laboratory Science, Nnamdi Azikiwe University Awka.
I have acquired direct hands-on experience with SARS Coronavirus and currently involved in SARS Coronavirus-2 (COVID-19) drug development, analytical development, in-vitro molecular diagnostic, cell-based assay and immuno-assay development as well as antiviral development, Maintenance of cell lines and cell cultures, Gene cloning and construction of recombinant viruses. molecular detection of Lassa fever virus, Phenotypic mixing of various viruses, antigenicity and immunogenicity testing of candidate HIV vaccines constructed from HIV .MPER, Virus neutralization assays. I have also worked with Dengue Virus, Chikungunya Virus, SARS Coronavirus, Influenza Virus, Respiratory Syncytial Virus, HIV. , polio and entero viruses, I have expertise in Infectious Diseases, Plaque Assay, TCID50, Virus Neutralization, ELISA, Nucleic Acid & amp; Antigen/Antibody detection assays, Immunofluorescence, Virus propagation/purification and characterization, Protein expression/purification, Process Development, Antibody production/Purification, RT-PCR, qRT-PCR, and Multiplex-PCR, waste water based epidemiology, oncovirology and neurovirology, plasmid manipulation and construction of artificial viruses. I also have extensive Biosafety Level 2+ Biosafety Level 3 work experience.
I have been able to conduct some research to determine the antigenicity of the membrane proximal external region (MPER) against some broadly neutralizing antibodies. This was performed in plasma samples from HIV discordant couples at the Chantal Biya International Reference center for prevention and management of HIV/AIDS in Yaounde Cameroun (Yet to be published). I successfully performed antigenicity testing of Membrane Proximal External Region(MPER) antigen using various immunogen associated immunoglobulins such as IgG subclasses ; IgG1, IgG2, IgG3, IgG4 and Immunoglobulin Isotpes i.e. IgG, IgGM, IgA. The Objective of the Immunogenicity tests was to determine the binding efficacy of different MPER associated immunogens in plasma (of study participants) to the MPER antigen. Since the MPER is one of the promising targets for HIV vaccine, positive results from my assay indicates the presence of neutralising antibodies in plasma of study participants. Our results therefore confirmed the MPER antigen as a potential vaccine target for HIV vaccine development This research ignited my interest to conduct more studies aimed at increasing the antigenicity of the MPER region of the HIV virus. In the course of my research, we engineered a coliphage virus and tried exposing more epitopes on the HIV MPER. This was in attempt to improve the antigenicity of the MPER region. Improving the antigenicity of the recombinant QβMPER phages will allow us to efficiently detect anti- MPER specific antibodies. In addition, I am also working on HBV immune responses in seronegative partners of HBV discordant couples. My research also tries to determine possible HBV genotypes associated with HBV discordant couples (unpublished). In addition, I study herbal extracts to determine their antiviral properties especially SARS-CoV-2.
